An investigation is carried out to determine the factors needed for photosynthesis using a healthy, green-leaved Pelargonium plant.

Before starting the investigation, the plant is kept in a dark room for 72 hours to destarch it.

Following this, two leaves on the plant are prepared as follows:

  • Leaf P: A circular disc of opaque black card is clipped securely over both the upper and lower surfaces of the middle of the leaf. The rest of the leaf remains uncovered and exposed to the air.
  • Leaf Q: The entire leaf is sealed inside a transparent, airtight plastic bag containing a small beaker of sodium hydroxide solution (which absorbs carbon dioxide, CO2\text{CO}_2CO2​).

The plant is then placed under an intensive LED grow lamp for 18 hours. Both leaves are then detached and tested for the presence of starch using iodine solution.
